Francisella tularensis is defined by the Centers for Disease Control and Prevention as a Category A bioterrorism agent due to its low infectious dose, high mortality rate, ease of aerosolization. Given there no licensed tularemia vaccine in United States possible development release antibiotic-resistant F. strains, an urgent need new treatments against this bacterium. We determined phenoxazine dye resazurin (Rz) exhibits antimicrobial activity other gram-negative bacteria. The mode action compound not understood, but potential targets Rz were identified throughput screen resistant isolates. Ninety-three percent Rz-resistant (Rzr) isolates sequenced contained mutations within coding regions FTL_0421 (lpnA), FTL_0895, FTL_1504 (katG). To confirm mutation lpnA was contributing resistance, we introduced wild-type copy into Rzr1 then assessed susceptibility resulting complemented strain (Rzr1/pABST-lpnA). minimum inhibitory concentration (MIC) Rzr1/pABST-lpnA similar, therefore, wanted restoring expression LpnA via Western blot analysis. As expected, expressed LVS genetic mutation.
